Cellecta’s first Single-module Human Genome-Wide CRISPR Knockout Library was designed to be small enough (~80,000 constructs) to enable convenient genome-wide “dropout viability” screens with most standard human cell modules. With 4 sgRNAs targeting each gene, all of which have been designed using the criteria for effective guides described by Doench, et al. and containing the Cellecta-designed HEAT sgRNA tracrRNA structure which we have proven generates more robust screening results than the standard sgRNA design, the library has deep enough gene coverage to generate efficient results.

The new Knockout-B library (Catalog# KOHGW-80K-B-xx) also has 4 sgRNA to each of the ~19,000 protein-coding genes
It is intended as a complement to the original 80,000-construct library (Catalog# KOHGW-80K-xx).
None of the sgRNA in the Knockout-B Library overlap with the original Human Genome-Wide CRISPR 80K Knockout Library.
The Knockout-B library may be run as an alternative to the original single-module library or added to the original library for researchers who want to run a screen with 8 guides for each gene.
The Knockout-B library is available with RFP and Puro markers.
Both libraries were cloned in the same vector--pRSG16-U6-(-sg)-UbiC-RFP-puro.
