Genetic Profiling and Functional Screening for Drug Target and Biomarker Discovery
Pooled libraries of heterogenous lentiviral constructs have proven to be a highly effective approach to individually label cells in a target population with cell-specific barcodes and/or other genetic effectors, such as CRISPR sgRNA targeting different genes. The transduced populations can then be analyzed by NGS to identify sub-sets of cells with specific traits or phenotypic responses which indicate the essentiality of particular genes or activation of certain pathways. In contrast, NGS analysis of targeted multiplex RT-PCR from cell and tissue samples, even down to single cell levels, enables parallel analysis of differences in gene activation across and within cell populations as a result of perturbation or selection. This presentation will discuss how these approaches can be used to gain a more comprehensive understanding of relevant and critical genetic pathways in cell models and diseased tissues.
