A major goal of my lab is to better characterize antigen specific T cell responses in the context of both infectious disease and cancer.  While the lab has historically used peptide-MHC multimers to identify and profile antigen specific cells, we have transitioned increasingly into using TCR sequencing-based approaches to achieve this goal.  I will describe an approach for using TCR sequence analysis to catalogue, track and phenotypically profile HIV vaccine specific T cell responses, as well as our recent efforts to predict colorectal cancer recurrence based on longitudinal TCR sequencing.
