DriverMap™ Cell-Free Adaptive Immune Receptor (cfAIR) RNA & DNA Repertoire Profiling Service
The DriverMap™ Cell-free Adaptive Immune Receptor (cfAIR) Repertoire Profiling Service profiles all TCR and BCR CDR3 regions from cell-free RNA and DNA in human plasma. Send us frozen plasma, and we purify the cell-free nucleic acids, perform library prep, sequence the libraries, and deliver a comprehensive analysis report — no biopsy, no dealing with live cells, and no in-house NGS workflow required.
Liquid-biopsy TCR/BCR profiling from as little as 1 mL of plasma
Two data sets from one sample: cfRNA (lymphocyte activation) and cfDNA (lymphocyte turnover), separated by barcodes from a single sample, in the same workflow
Captures clonotypes from tumor-infiltrating and tissue-resident lymphocytes that blood-cell adaptive immune receptor (AIR) repertoire profiling does not show
Complete end-to-end service from plasma to report, with a turnaround of as little as 3 weeks
General Information
The DriverMap™ Cell-Free AIR service workflow is designed to obtain the clonotype repertoire:
Amplify the CDR3 region of all seven TCR and BCR chains (TRA, TRB, TRG, TRD, IGH, IGK, IGL) with short FR3–J amplicons of approximately 170 bp designed for fragmented cell-free templates.
Count template molecules and correct PCR and sequencing errors with Validator Barcodes (VBCs), for accurate detection of rare clonotypes present at as few as 1–10 copies.
Sample Requirements
Sample
Requirement
Plasma (required)
1–6 mL per sample; 4–5 mL recommended
Blood collection
K2 EDTA tubes (ACD-A or PPT tubes also accepted). Do not use blood-stabilizing tubes (for example, Streck Nucleic Acid BCT or PAXgene Blood ccfDNA).
Plasma preparation
Process within 2–4 hours of collection. Double spin: 2,500 × g, 15 min, 20 °C, no brake (twice). Freeze aliquots at –80 °C. Avoid freeze–thaw cycles.
Matched control (recommended)
Buffy coat or PBMC (or 100 ng purified RNA); FFPE tumor tissue (or 500–1,000 ng purified RNA)
Shipping
Frozen on dry ice
Some Applications
Cancer Immunotherapy Monitoring Track tumor-reactive TCR and BCR clonotypes in plasma before and during checkpoint-inhibitor therapy, without the need for invasive biopsy.
Lymphoid Malignancy Detection Detect and follow malignant B-cell and T-cell clones in plasma cfDNA and cfRNA.
Biobanked Plasma Characterization Add immune repertoire data to further characterize plasma already collected for ctDNA or other liquid-biopsy studies.
