Product Information

Obtain the full-length sequence of both chains of T-cell (TCR) or B-cell receptors (BCR) from flow-sorted or pipetted T- or B-cells with the DriverMap Single-Cell Adaptive Immune Receptor (scAIR) TCR-BCR assays. These consist of a variety of easy-to-use, 96-well, plate-based RT-PCR kit configurations 

  • Full-length sequences of both TCR alpha/beta or BCR IGH/IGK/IGL receptor chains: With the DriverMap scAIR Chain Pairing Reagent Kit, you obtain complete coding region sequences for human, mouse, and mouse/human hybrid T-cell (TRA-TRB) or B-cell(IGH-IGK-IGL) receptors.
    • Additionally, with the DriverMap scAIR Immunophenotyping Kits, you obtain immunophenotyping information from 36 T-cell or 30 B-cell expression marker genes for further info on cell state
  • Simple, streamlined protocol: Sort cells into an indexed 96-well plate with RT primers, hybridize, then pool samples into a single reaction tube. Perform RT-PCR and next-generation sequencing (NGS), scTCR-Seq or scBCR-Seq. No RNA extraction, specialized equipment, or complex barcoding required.
  • Characterize full receptors from single cells or 1000s of cells: Two workflow options--the first for 96 flow-sorted individual cells per plate; and the second for up to ~10,000 cells (~100cells/well) to thoroughly characterize each receptor in small numbers of cells, or extract large numbers of abundant receptor chain pairs in larger cell populations.
  • Cost-effective protocols: Economical price point, starting from approximately 10 cents per cell.
  • Integrated bioinformatics analysis pipeline with MiXCR software on Platforma (MiLaboratories, Inc.) to process FASTQ files, obtain clonotype alignment and chain-pair information, and, optionally, for human samples, analyze expression of key marker genes that show the immunophenotype of each cell. Other open-source tools may also be used for further data processing.

DriverMap scAIR TCR/BCR Reagent Kits include all the enzymes, reagents, and primers to go from samples to NGS-ready libraries. A separate Universal Unique Dual-Index NGS plate is required for next-generation sequencing (NGS) run: scTCR-Seq or scBCR-Seq.

Cellecta DriverMap Single-Cell TCR-BCR Profiling Assay Workflow

Fig 2: DriverMap scAIR Chain Pairing Assay Workflow for TCR/BCR paired-chain analysis from flow-sorted or pipetted single cells or multiple cells per well.  The hybridization plate contains a combination of forward gene-specific primers (Fwd GSPs) targeting the FR1 region and barcoded reverse gene-specific primers (Rev GSPs) targeting the C region of TCR/BCRs. RevGSPs have 96 cell/well-specific barcodes (Cell-BC) and validator barcodes (VBCs) to facilitate quantitative analysis of paired receptor chains. Before cDNA synthesis, all cells may be pooled into a single tube for easy handling. Anchor 1 and Anchor 2 are sequences of universal primers used in the first amplification step. P5-UD1-Anchor1 and P7-UD2-Anchor2 are primers with Illumina UD indexes used in the second PCR step. The resulting NGS data provide chain-pairing information and clonotype abundance levels for each gene. 

 

Workflow Requirements 

Each DriverMap™ scAIR TCR-BCR Assay Reagent Kit includes all specialized enzymes, primers, and reagents required for library preparation; however, to run the libraries on Illumina® next-generation sequencing (NGS) platforms, a compatibleUniversal Unique Dual Index (UDI) NGS Index Platemust be purchased separately as a required component of the workflow.
